Behom trávenia majú významnejšiu úlohu chemické enzýmy, produkované mikroorganizmami. Mikrobióm tvoria najmä baktérie, ale aj nálevníky, archaea, anaeróbne huby a v menšej miere aj bakteriofágy. Cieľom tejto bakalárskej práce bolo sledovať zmeny v zastúpení mikroorganizmov v ruminálnej tekutine počas kultivácie. Do bioreaktora bolo pridávané krmivo a v priebehu fermentácie boli odoberané vzorky bachorovej tekutiny. Bola nameraná koncentrácia celého bachorového obsahu a koncentrácia bakteriálnych kmeňov Bacteroidetes a Firmicutes. Stabilita v bioreaktore bola vyhodnotená pomocou RT -qPCR analýzy, ktorá zaznamenala stálu koncentráciu mikroorganizmov počas kultivácie. | 520 | 2 | 9 | |a Microbiome stability in the rumen of the ruminants is the key condition for proper digestion and processing of received nourishment. Food in the rumen is mechanically and chemically digested. During digestion, chemical enzymes produced by microorganisms have more important role. Microbiome consists mainly of bacteria, but also of archaea, anaerobic fungi and in smaller extent also of bacteriophages. The aim of this Bachelor thesis was to monitor changes in representation of microorganisms in the ruminal fluid during cultivation. The feed was being added into the bioreactor and during the fermentation were collected samples of ruminal fluid. The concentration of whole ruminal content and the concentration of bacterial phyla Bacteriodetes and Firmicutes were measured. Stability in bioreactor was evaluated by RT-qPCR analysis, which recorded stable concentration of microorganisms during cultivation.
