Charakterizace lektinu BC2L-C z podmíněného patogenu Burkholderia cenocepacia
BC2L-C je rozpustný lektín pochádzajúci z podmieneného patogénu Burkholderia cenocepacia, ktorý spôsobuje život-ohrozujúce infekcie u pacientov s cystickou fibrózou. Tento lektín sa skladá z dvoch domén: N-terminálnej domény, ktorá nemá doposiaľ popísaného sekvenčného homológa a C-terminálnej domény, ktorá je sekvenčne podobná lektínu BCLA. Táto práca je zameraná na skúmanie termálnych stabilít lektínu BC2L-C a jeho domén a určenie oligomérnych vlastností BC2L-C. Pomocou CD spektrometrie a diferenčnej skenovacej mikrokalorimetrie bola určená denaturačná teplota Tm lektínu BC2L-C a jeho domén. Taktiež bol pozorovaný vplyv cukrov (D-manózy a L-fukózy) a prostredie glycerolu na ich teplotnú stabilitu. Sekundárna štruktúra BC2L-C C-terminálnej domény a BC2L-C N-terminálnej domény bola určená pomocou CD spektroskopie a v obidvoch prípadoch bola potvrdená štruktúra β-skladaného listu. Oligomerizačné vlastnosti celého lektínu BC2L-C boli skúmané pomocou gélovej permeačnej chromatografie. Lektín po purifikácií s použitím elučného pufru s obsahom EDTA ovplyvňovala oligomerizáciu proteínu a vyskytoval sa najmä vo forme monoméru. |% cze | |
| 520 | 2 | 9 | |a BC2L-C is a soluble lectin from an oportunistic pathogen Burkholderia cenocepacia causing infections in patients with cystic fibrosis. This lectin consists of two domains:N- terminal domain, which has not any sequence homologue yet, and C-terminal domain with a sequence similarity to the lectin BCLA. This study is focused on exploring the thermal stability of the lectin BC2L-C and its domains and the oligomeric characteristics of BC2L-C. Using CD spectroscopy and differential scanning microcalorimetry, the temperature of denaturation Tm of BC2L-C and its domains were determined. The influence of carbohydrates (L-fucose and D-mannose) and of glycerol was described due to its thermal stability. The secondary structure of BC2L-C N-terminal domain and C-terminal domain was determined by CD spectroscopy as β - sheets. The oligomerization of the lectin was evaluated using gel permeation chromatography. The lectin after purification with elution buffer containing mannose was mainly in hexameric structure with a contribution of trimeric structure. However, the purification using EDTA as an elution buffer influences the oligomerization of protein and it consists mainly of monomers. |9 eng |
