Interakcie jedného z najčastejšie sa vyskytujúcich mutantných proteínov - R175H s izolovanými centrálnymi doménami p63 a p73 boli skúmané imunoprecipitačnou technikou in vitro. Získané výsledky boli v súlade so závermi zverejnenými v literatúre, tie však pochádzali z experimentov s celými proteínmi. Ďalej bol skúmaný vplyv mutantných proteínov (G245S, R248W, R249S) na sekvenčne špecifickú väzbu izolovaných centrálnych domén p63 a p73 na DNA pomocou elektroforézy z agorózovom géle. Podrobnejšie znalosti o spôsobe akým dochádza k útlmu nádor supresívnych vlastností členov rodiny v prítomnosti mutantnej p53 by mohli napomôcť presnejšiemu diagnostikovaniu a postupom v proti-nádorovej liečbe. |% cze | |
| 520 | 2 | 9 | |a In this thesis the protein-protein interactions between purified mutant p53 and purified protein core domains p63 and p73 were analyzed. Interactions were studied by imunoprecipitation in vitro. One of the most abundant p53 mutants - R175H and the isolated core domains of p63 a p73 were investigated. Obtained results were in accord with published data, unlike studied isolated core domains, describing interactions utilizing full-length p63 and p73 proteins. Furthermore, the influence of mutant proteins (G245S, R248W a R249S) on sequence-specific binding ability of p63 and p73 isolated core domains were analyzed by electro-mobility shift assays in agarose gels. Understanding molecular basis of mutant p53 impact on tumour suppressor function of p63 and p73 could have a great significance in anti-cancer therapy. |9 eng |
