RNA motívy nazývané Kink turny (K-turn) sú asymetrické vnútorné loopy, charakteristické ostrým zahnutím fosfodiesterového skeletu. K-turny sa často nachádzajú v blízkosti kľúčových miest ribozómu. Uskutočnili sme rozsiahlu, počítačovú, štruktúrno-dynamickú štúdiu vybraných K-turn motívov za prítomnosti explicitného solventu. Použitím súčasných výpočtových techník dopĺňame znalosti o štruktúrnom a dynamickom správaní týchto RNA na atomárnej úrovni, a výrazne tak obohacujeme dostupné informácie o molekulových interakciách v ribozóme. Zistili sme, že RNA Kink turn motívy sú konštruované ako flexibilné, molekulové nano-kĺby, schopné umožniť biologicky významné pohyby rôznych častí ribozómu počas jednotlivých kroko. |9 slo |
| 520 | 2 | 9 | |a In presented PhD thesis I have carried out a wide range of investigations of structural dynamics and molecular interactions of rRNA molecules (e.g. RNA Kink turn motifs, GTPase associated center rRNA, Helix 44 from 16S rRNA), using a variety of Molecular Dynamics (MD) methods. RNA motifs called Kink turns (K-turn) are asymmetrical internal loops, characterized by sharp bend of phosphodiester backbone. K-turns are often localized adjacent to the key functional sites in the ribosome. Extensive computational structure-dynamical study of selected K-turn motifs (in the presence of explicit solvent) was performed. Using current state-of-art computational techniques, I complemented the knowledge about the structural and dynamical behavior of these RNAs at atomic level, and significantly enriched the available information about molecular interactions in the ribosome. |9 eng |
